General Description

Methacholine Chloride is a synthetic choline ester of acetic acid classified as a muscarinic cholinergic agonist. It is resistant to acetylcholinesterase hydrolysis due to its beta-methyl substitution.

Mechanism of Action

Methacholine directly stimulates muscarinic acetylcholine receptors, primarily M3 receptors on bronchial smooth muscle, causing bronchoconstriction. In normal individuals this effect is minimal, but it is exaggerated in asthma patients.

Application

Used in bronchoprovocation testing. Methacholine Chloride is indicated for diagnosing asthma, assessing airway hyperresponsiveness, and evaluating the efficacy of anti-asthma therapy.

Why is Methacholine useful for diagnosing asthma?

Methacholine produces dose-dependent bronchoconstriction in asthmatic patients at concentrations that have minimal effect on non-asthmatic individuals, providing a sensitive diagnostic marker for airway hyperresponsiveness.
