General Description
Paramethasone acetate is a synthetic glucocorticoid with potent anti‑inflammatory activity and minimal mineralocorticoid effects. It is available as oral tablets for systemic use and has a longer duration of action than prednisone. The drug is a fluorinated corticosteroid, structurally related to dexamethasone, but is now rarely prescribed.
Mechanism of Action
Paramethasone binds to cytoplasmic glucocorticoid receptors, and the complex translocates to the nucleus to regulate gene transcription. It suppresses the production of pro‑inflammatory cytokines (IL‑1, IL‑6, TNF‑alpha), reduces phospholipase A2 expression, and inhibits NF‑κB activation. These actions result in profound immunosuppression, anti‑inflammation, and stabilization of lysosomal membranes.
Application
Paramethasone was indicated for a wide range of inflammatory and autoimmune conditions, including rheumatoid arthritis, asthma, allergic disorders, and dermatologic diseases. It was also used as adjunctive therapy in certain cancers and for cerebral edema.
