General Description

Lypressin is a synthetic octapeptide analog of vasopressin in which arginine at position 8 is replaced by lysine. It retains antidiuretic activity while exhibiting reduced pressor effects compared to native vasopressin.

Mechanism of Action

Lypressin activates V2 receptors in renal collecting duct cells, stimulating adenylate cyclase and increasing cyclic AMP, which promotes aquaporin-2 insertion into the apical membrane, enhancing water reabsorption.

Application

Used in the treatment of central diabetes insipidus. Lypressin is indicated for vasopressin-deficient polyuria and polydipsia resulting from hypothalamic-pituitary damage.

How does Lypressin differ from desmopressin in diabetes insipidus management?

Lypressin retains some V1 receptor activity causing mild vasoconstriction, while desmopressin is more selective for V2 receptors, offering a wider therapeutic window.
