General Description
Ibuprofen lysine is the lysine salt of the nonsteroidal anti‑inflammatory drug ibuprofen, designed to enhance water solubility and improve the onset of absorption. The molecule pairs the propionic acid derivative ibuprofen with the basic amino acid L‑lysine. The lysine ion neutralizes the carboxyl group of ibuprofen, forming a highly soluble salt without altering the pharmacological activity of the ibuprofen moiety.
Mechanism of Action
Once absorbed, the salt dissociates and the ibuprofen component reversibly inhibits cyclooxygenase (COX‑1 and COX‑2) enzymes, blocking the synthesis of prostaglandins involved in pain, fever, and inflammation. The lysine moiety does not contribute to the pharmacodynamic effect, but the increased solubility results in faster dissolution and possibly more rapid absorption compared to ibuprofen free acid. The drug retains the same target selectivity profile.
Application
Ibuprofen lysine is indicated for the management of mild to moderate pain, fever, and inflammation, particularly in situations where rapid onset is desired. It is also used for patent ductus arteriosus in preterm neonates (intravenous formulation), relying on the same mechanism of prostaglandin inhibition to constrict the ductus.

Does Ibuprofen Lysine require protection from moisture during storage?

Yes, it is hygroscopic. The lysine salt readily absorbs moisture, leading to caking and potential degradation. Store in tightly sealed containers with desiccant.

What is the recommended storage temperature for Ibuprofen Lysine?

Store at controlled room temperature (15-25°C). Avoid excessive heat above 30°C, which can accelerate oxidative degradation and racemization.

Is Ibuprofen Lysine stable in solution for intravenous administration?

Aqueous solutions are stable when protected from light and stored at room temperature for up to 24 hours.

How is the impurity 4-isobutylacetophenone (an oxidative degradation product) monitored?

This degradation product is quantified using a stability-indicating HPLC method with UV detection, ensuring it remains below ICH limits.
