General Description
Clonidine is a synthetic imidazoline derivative with a dichlorophenyl group, acting as a centrally acting alpha-2 adrenergic agonist. Its chemical structure is 2,6-dichloro-N-(2-imidazolidinylidene)aniline. Clonidine is a white to off-white crystalline powder, soluble in water and alcohol. The molecule has a high affinity for alpha-2 adrenergic receptors.
Mechanism of Action
Clonidine acts as a selective agonist at alpha-2 adrenergic receptors in the brainstem (vasomotor center), reducing sympathetic outflow from the central nervous system. This leads to a decrease in peripheral vascular resistance, heart rate, and blood pressure. The drug also has a weak action at imidazoline (I1) receptors, contributing to its antihypertensive effect.
Application
Clonidine is indicated for the treatment of hypertension, particularly in patients who are not controlled on other antihypertensive agents. It is also used for the management of attention-deficit/hyperactivity disorder (ADHD) and for the treatment of opioid withdrawal. The drug is effective in reducing the symptoms of anxiety and panic disorders.

Does Clonidine require protection from light during long-term storage?

Yes, it is photosensitive. UV exposure can cause photodegradation and discoloration of the imidazoline ring. Store in light-resistant containers, preferably amber glass.

What is the recommended storage temperature for Clonidine?

Store at controlled room temperature (15-25°C). Avoid excessive heat above 30°C, which can accelerate degradation of the dichlorophenyl ring.

Is Clonidine stable in transdermal patch and oral formulations?

Yes, when formulated with standard excipients and protected from light.

How is the impurity clonidine N-oxide (an oxidative degradation product) monitored?

This degradation product is quantified using a stability-indicating HPLC method, ensuring it remains within pharmacopoeial limits.
