General Description
Betamethasone Acetate is the 21-acetate ester of betamethasone, a synthetic fluorinated glucocorticoid. The acetate ester provides reduced aqueous solubility compared to betamethasone sodium phosphate, resulting in a depot effect that extends the duration of glucocorticoid activity at the site of administration.
Mechanism of Action
Betamethasone Acetate binds to intracellular glucocorticoid receptors, and the receptor-ligand complex translocates to the nucleus to modulate gene transcription. It suppresses the expression of pro-inflammatory cytokines (IL-1, IL-6, TNF-alpha), adhesion molecules, and cyclooxygenase-2, while upregulating anti-inflammatory mediators.
Application
Indicated for the treatment of inflammatory and pruritic dermatoses. Betamethasone Acetate is a synthetic fluorinated glucocorticoid with a depot effect from its acetate ester, providing prolonged anti-inflammatory activity at the injection site for localized steroid-responsive conditions.
Betamethasone acetate-loaded polyvinyl alcohol nanospheres were prepared by oil-in-water emulsion method followed by different cycles of freeze-thawing process. Transmission electron microscopy confirmed successful fabrication of nanospheres. The freeze-thawing treatment physically cross-linked the PVA matrix, and increasing the number of freeze-thaw cycles from 1 to 4 significantly decreased the rate of drug release from the nanospheres. These biocompatible nanospheres achieved sustained drug release for more than four months. MTT assay on human chondrosarcoma SW 1353 cells confirmed the biocompatibility of the nanospheres.
