General Description
Vidarabine Monophosphate (ara-A monophosphate) is the monophosphate ester of vidarabine (ara-A), an analog of the natural nucleoside adenosine in which the ribose sugar is replaced by D-arabinose. The monophosphate form is the initial intracellular metabolite of vidarabine, further phosphorylated to the active triphosphate (ara-ATP) that inhibits viral DNA polymerase.
Mechanism of Action
Vidarabine Monophosphate is an antimetabolite that functions through its triphosphate metabolite (ara-ATP). Vidarabine monophosphate is further phosphorylated by cellular kinases to ara-ATP, which competitively inhibits viral DNA polymerase with higher affinity than host cell DNA polymerases. It is also incorporated into viral DNA, where it causes chain termination. Unlike acyclovir, vidarabine does not require virus-specific kinase activation, making it effective against a broader range of DNA viruses.
