General Description
Tripelennamine Hydrochloride is a first-generation antihistamine (H1 receptor antagonist) of the ethylenediamine class, notable for its relatively rapid onset and moderate duration of action. The hydrochloride salt provides enhanced aqueous solubility and is suitable for both oral and topical (dermal) administration. It has been used clinically since the mid-20th century for allergy management.
Mechanism of Action
Tripelennamine Hydrochloride competitively antagonizes histamine H1 receptors on smooth muscle cells, endothelial cells, and sensory nerve endings, blocking histamine-mediated vasodilation, increased vascular permeability, edema formation, and pruritus. Its ethylenediamine structure confers a degree of anticholinergic activity that contributes to drying effects on respiratory secretions.
Application
Tripelennamine Hydrochloride is indicated for the symptomatic treatment of allergic conditions including allergic rhinitis, hay fever, urticaria, and allergic skin reactions.
