General Description
Triamcinolone Diacetate is a synthetic glucocorticoid corticosteroid administered primarily via intra-articular, intralesional, or intramuscular routes as an injectable suspension. The diacetate ester provides a microcrystalline suspension formulation that allows for extended depot-like release from injection sites, prolonging the duration of anti-inflammatory action compared to aqueous solutions of the base steroid.
Mechanism of Action
Triamcinolone Diacetate exerts potent anti-inflammatory and immunosuppressive effects by binding to and activating the glucocorticoid receptor (GR), leading to the regulation of genes involved in inflammatory and immune responses. It inhibits phospholipase A2 through lipocortin-1 induction, suppresses the synthesis of inflammatory prostaglandins and leukotrienes, reduces the synthesis and release of inflammatory cytokines, and suppresses the proliferation and activity of multiple immune cell populations.
Application
Triamcinolone Diacetate is indicated for intra-articular, intrabursal, tendon sheath, and intralesional administration in the treatment of rheumatoid arthritis, osteoarthritis, acute gouty arthritis, and various inflammatory conditions of joints and soft tissues. It is also used for the treatment of allergic and inflammatory conditions of the skin, respiratory tract, and eyes, and for certain adrenal insufficiency states.
