General Description

Pirbuterol Acetate is a selective beta-2 adrenergic receptor agonist bronchodilator with a unique pyridine heterocyclic structure. Unlike traditional catecholamine derivatives, its non-catechol structure confers resistance to COMT-mediated degradation, providing a longer duration of action and allowing oral administration in addition to inhalation. The acetate salt is used in inhalation aerosol formulations.

Mechanism of Action

Pirbuterol Acetate directly activates beta-2 adrenergic receptors on bronchial smooth muscle cells, triggering a Gs protein-coupled signaling cascade that activates adenylate cyclase, raises intracellular cAMP levels, and causes smooth muscle relaxation and bronchodilation. Its non-catechol structure renders it resistant to inactivation by catechol-O-methyltransferase (COMT), extending its duration of action. The selectivity for beta-2 over beta-1 receptors minimizes cardiac side effects at recommended doses.
