General Description

Pentetate Zinc Trisodium (Zn-DTPA) is the zinc salt of diethylenetriaminepentaacetic acid (DTPA), a chelating agent structurally related to EDTA but with enhanced capacity for binding actinides and transuranic elements. As the zinc salt, it is used primarily as an alternative to the calcium salt (Ca-DTPA) in the decorporation of internally deposited radioactive and heavy metal contaminants.

Mechanism of Action

Pentetate Zinc Trisodium acts by chelating and mobilizing internally deposited radionuclides and heavy metals from body tissues and fluids through competitive binding. Zinc-DTPA is particularly effective for chelating transuranic elements such as plutonium, americium, and curium, forming stable, water-soluble complexes that are rapidly excreted in the urine. The zinc cation is released and reabsorbed, providing essential zinc without depleting calcium.

Pentetate Zinc Trisodium is indicated for the treatment of internal contamination with plutonium, americium, and curium in cases of radiation exposure or nuclear accidents.

Pentetate zinc trisodium (Zn-DTPA) is a chelating agent used to treat internal contamination with transuranium elements (plutonium, americium, curium). It forms stable, soluble chelates with these radioactive metals by exchanging zinc for a metal with greater binding capacity. The radioactive chelate is then excreted by glomerular filtration into urine.
Zn-DTPA is poorly absorbed orally; intravenous or inhalation administration is used. Following IV administration, it rapidly distributes throughout extracellular fluid without cellular penetration, undergoes minimal metabolism, and is eliminated unchanged by glomerular filtration. Zn-DTPA is preferred for maintenance chelation therapy after the initial Ca-DTPA dose because it causes less depletion of endogenous trace metals (zinc, magnesium, manganese). Animal studies show no teratogenicity.
