General Description
Methixene Hydrochloride is an anticholinergic and antispasmodic agent of the thioxanthene class, structurally related to the antipsychotic drug thiothixene but designed for peripheral anticholinergic effects. It competitively antagonizes muscarinic acetylcholine receptors on gastrointestinal smooth muscle, reducing acetylcholine-mediated peristaltic contractions and gastrointestinal spasms, while also exerting mild antihistamine effects.
Mechanism of Action
Methixene Hydrochloride acts as a competitive antagonist at muscarinic acetylcholine receptors (M1, M2, M3 subtypes) in the smooth muscle of the gastrointestinal tract, urinary bladder, and other organs. By blocking the parasympathetic cholinergic transmission that drives smooth muscle contractions, it reduces gastrointestinal motility, intestinal spasms, and visceral pain associated with functional bowel disorders. Its antihistamine properties provide additional antipruritic and mild sedative effects.
Application
Methixene Hydrochloride is indicated for the symptomatic treatment of gastrointestinal spasm, irritable bowel syndrome (IBS), intestinal colic, and other functional gastrointestinal disorders characterized by hypermotility and visceral hypersensitivity. It may also be used in the management of urinary frequency and urgency associated with overactive bladder.
