General Description

Hydrocodone is a semi-synthetic opioid analgesic, a derivative of codeine with a ketone group, belonging to the class of phenanthrenes. Its chemical structure is 4,5α-epoxy-3-methoxy-17-methylmorphinan-6-one. Hydrocodone is a white to off-white crystalline powder, practically insoluble in water but soluble in organic solvents. It is a potent mu-opioid receptor agonist.

Mechanism of Action

Hydrocodone binds to mu-opioid receptors in the central nervous system, activating the G-protein coupled receptor and inhibiting the release of neurotransmitters, such as substance P and glutamate. This leads to a reduction in the transmission of pain signals and an increase in the pain threshold. The drug also produces euphoria and sedation, which contribute to its abuse potential.

Application

Hydrocodone is indicated for the treatment of moderate to severe pain, including acute and chronic pain. It is also used as an antitussive for the relief of cough. The drug is available in combination with other analgesics, such as acetaminophen and ibuprofen.
