General Description
Glutethimide is a hypnotic and sedative that was introduced in 1954 as a potential non-barbiturate alternative for the treatment of insomnia.
Mechanism of Action
The exact mechanism of action has not been fully elucidated, but it is believed to function as a GABA agonist, which helps induce sedation. By potentiating GABAergic transmission in the central nervous system, it produces central nervous system depression. Additionally, it induces CYP2D6, which can alter the metabolism of co-administered drugs that are substrates of this enzyme.
Application
Glutethimide was originally indicated for the treatment of insomnia, but its use has now been largely superseded by other drugs with better safety profiles. Its historical use has also included the treatment of essential tremor and intention tremor, with evidence suggesting efficacy.
