General Description
Ethynodiol Diacetate is the 3,17-diacetate ester of ethynodiol, a synthetic progestogen of the 19-nor steroid class. It is a prodrug that undergoes hydrolysis to ethynodiol, which is subsequently metabolized to norethisterone (norethindrone), the principal active progestogen.
Mechanism of Action
After ester hydrolysis and metabolic conversion to norethisterone, ethynodiol diacetate activates progesterone receptors to suppress LH surges, reduce endometrial proliferation, increase cervical mucus viscosity, and modulate tubal motility. It also exhibits weak androgenic activity through its norethisterone metabolite.
Application
Indicated as a progestogenic component in combined oral contraceptives. Ethynodiol Diacetate is a 19-nor steroid prodrug that undergoes sequential deacetylation and metabolic conversion to norethisterone, the principal active progestogen responsible for ovulation suppression and contraceptive efficacy.
