The solubility of Erythromycin Ethylsuccinate in six organic solvents (tetrahydrofuran, acetone, acetonitrile, hexane, ethanol, 2-propanol) was measured at 288.15–323.15 K using gravimetry. Solubility increased with temperature (endothermic dissolution). Data were correlated with four models; modified Apelblat gave the best fit. Mixing thermodynamics from NRTL showed spontaneous dissolution. Hansen solubility parameters explained the observed order: hexane < acetonitrile < ethanol < 2-propanol < acetone < tetrahydrofuran.

Erythromycin ethylsuccinate was formulated into solid lipid nanoparticles using heating emulsification and homogenization methods to enhance its aqueous solubility. Formula 1 achieved an encapsulation efficiency of 85.688 percent with a particle size of 398.9 nm, polydispersity index of 0.3895, and zeta potential of -17.45 mV. The solubility of erythromycin ethylsuccinate in the solid lipid nanoparticle formulation was increased compared to the pure substance. Stability was determined by accelerated testing under extreme temperature and mechanical stress conditions, with pH decrease and particle precipitation serving as indicators of instability.
