General Description
Didanosine is a synthetic purine nucleoside analogue, a reverse transcriptase inhibitor, with a dideoxy structure. Its chemical structure is 2′,3′-dideoxyinosine. Didanosine is a white to off-white crystalline powder, soluble in water. It is a prodrug that is converted to the active metabolite, dideoxyadenosine triphosphate (ddATP), in the body.
Mechanism of Action
Didanosine is phosphorylated intracellularly to the active triphosphate, dideoxyadenosine triphosphate (ddATP), which competes with the natural substrate deoxyadenosine triphosphate (dATP) for incorporation into the viral DNA. Once incorporated, it acts as a chain terminator, preventing further elongation of the DNA strand. This inhibits the replication of HIV-1.
Application
Didanosine is indicated for the treatment of HIV-1 infection in combination with other antiretroviral agents. It is used in patients who are treatment-naïve or who have experienced treatment failure with other nucleoside analogues.

Does Didanosine require protection from acid and moisture during storage?

Yes, it is acid-labile and hygroscopic. The purine ring is susceptible to hydrolysis in the presence of moisture and acidic conditions. Store in tightly sealed containers with desiccant.
