General Description
Cefpiramide Sodium is a third-generation cephalosporin distinguished by an exceptionally long elimination half-life and dual hepatobiliary and renal excretion pathways, supporting once-daily dosing.
Mechanism of Action
Cefpiramide binds to penicillin-binding proteins in the bacterial cell wall, inhibiting transpeptidation of peptidoglycan cross-links. This weakens the cell wall and activates autolytic enzymes, resulting in bactericidal lysis.
Application
Used in the treatment of serious bacterial infections. Cefpiramide Sodium is indicated for respiratory tract infections, urinary tract infections, biliary tract infections, peritonitis, and septicemia caused by susceptible organisms.
Cefpiramide sodium-loaded liposomes were prepared by reverse phase evaporation method to improve the pharmacokinetics and tissue distribution of the antibiotic. In mice receiving a single intravenous injection of 152 mg/kg of either liposomal cefpiramide or cefpiramide solution, the liposomal formulation showed 2.8-fold higher bioavailability and 4.5-fold longer mean residence time compared to the solution. The targeting efficiency ratio of lung to other tissues was significantly elevated for liposomal cefpiramide, with an intake rate of 2.97 for the lung being the highest among tested tissues. The liposomes demonstrated a promising sustained-release and drug-targeting system for cefpiramide sodium.
