General Description
Carprofen is a synthetic nonsteroidal anti-inflammatory drug (NSAID) with a carbazole core and a propionic acid side chain. Its chemical structure is 2-(6-chloro-9H-carbazol-2-yl)propanoic acid. Carprofen is a white to off-white powder, practically insoluble in water but soluble in organic solvents. The molecule is a chiral compound, and the S-enantiomer is the more active form.
Mechanism of Action
Carprofen acts as a selective inhibitor of cyclooxygenase-2 (COX-2), with less inhibition of COX-1, reducing the synthesis of prostaglandins involved in inflammation and pain. This results in anti-inflammatory, analgesic, and antipyretic effects. The drug also has chondroprotective properties, inhibiting the degradation of cartilage.
Application
Carprofen is indicated for the management of pain and inflammation associated with osteoarthritis and other musculoskeletal disorders. It is also used for the control of postoperative pain. The drug is effective in reducing joint pain, swelling, and stiffness, improving mobility and quality of life.

In canine chondrocyte microcarrier cultures, IL‑1β stimulation increased PGE₂, IL‑6, IL‑8, and MCP‑1. Carprofen (NSAID) and a combination of avocado/soybean unsaponifiables, glucosamine, and chondroitin sulfate (ASU+GLU+CS) each suppressed these increases. The combination of carprofen with ASU+GLU+CS reduced PGE₂ more than either alone, though ASU+GLU+CS alone was more effective than carprofen against IL‑6, IL‑8, and MCP‑1. The findings suggest that adding ASU+GLU+CS to low‑dose carprofen could provide safe, effective joint pain management.

In oral squamous cell carcinoma (OSCC) patients, plasma CGRP levels correlated positively with pain severity and reduced overall survival. Increased CGRP in tumor-infiltrating nerves was linked to greater pain and lymph node metastasis. In an OSCC mouse model, both systemic and local administration of the CGRP receptor antagonist rimegepant attenuated facial mechanical hypersensitivity. Transcriptomic analysis revealed suppression of innate immune and neuroinflammatory pathways. CGRP receptor antagonism warrants further evaluation as an analgesic strategy in cancer pain.

Does Carprofen require protection from light during long-term storage?

Yes, it is photosensitive. UV exposure can cause photodegradation and discoloration of the carbazole ring. Store in light-resistant containers, preferably amber glass.
