General Description
Calcium Acetate is an organic calcium salt serving dual functions as a phosphate binder and calcium supplement. It is highly effective in controlling serum phosphorus levels in renal failure.
Mechanism of Action
Calcium Acetate dissociates in the intestine, where calcium ions bind dietary phosphate to form insoluble calcium phosphate that is excreted in feces. This reduces phosphorus absorption while simultaneously supplementing calcium.
Application
Used in the management of hyperphosphatemia. Calcium Acetate is indicated for serum phosphorus control in patients with end-stage renal disease on dialysis, and for hypocalcemia associated with chronic kidney disease.

A novel compound effervescent granule of calcium acetate was developed using wet granulation method with citric acid and sodium bicarbonate as effervescent base, optimized by orthogonal experiment. The formulation consisted of calcium acetate, citric acid, sodium bicarbonate, PEG6000, aspartame, PVP ethanol solution, lactose and vitamin D. The granules were evaluated for particle size, repose angle, pH value, calcium acetate content and effervescence time. In vivo studies demonstrated that the effervescent granules exhibited prominent bone-mass loss prevention and superior bone microarchitecture enhancement compared to other calcium preparations in Micro-CT analysis. This novel dosage form offers a promising approach for calcium supplementation with improved palatability and therapeutic efficacy.
