General Description
Betamethasone Benzoate is a synthetic corticosteroid ester combining betamethasone with benzoic acid, designed for enhanced topical bioavailability and penetration into the skin. The benzoate ester increases lipophilicity compared to the base betamethasone molecule, facilitating more efficient delivery across the stratum corneum. It is classified as a high-potency topical corticosteroid.
Mechanism of Action
Betamethasone Benzoate acts through glucocorticoid receptor (GR)-mediated modulation of gene transcription, inducing the synthesis of lipocortin-1 (annexin-1), which inhibits phospholipase A2 and thereby reduces the formation of arachidonic acid derivatives including prostaglandins and leukotrienes. It also suppresses the migration of polymorphonuclear leukocytes, reduces capillary permeability, and exerts antiproliferative effects on fibroblasts, collectively attenuating the inflammatory cascade in skin.
Application
Betamethasone Benzoate is indicated for the topical treatment of corticosteroid-responsive dermatoses including atopic dermatitis, contact dermatitis, psoriasis, seborrheic dermatitis, and lichen planus where high-potency anti-inflammatory action is required.
