General Description

2,4-Piperadinedione (also known as glutarimide or 2,4-diketopiperidine) is a six-membered saturated heterocycle featuring two carbonyl groups at the C-2 and C-4 positions. This diketopiperazine-like structure is the core scaffold of the glutarimide class of natural products and pharmaceuticals, which includes the immunosuppressant cyclosporine A and the antibiotic streptozocin. The piperidine-2,4-dione ring is a constrained analog of gamma-aminobutyric acid (GABA).

Mechanism of Action

2,4-Piperadinedione functions as a GABA-like scaffold and as a building block for the synthesis of glutarimide derivatives. The diketopiperidine ring can undergo N-alkylation, and the activated methylene at C-5 is subject to electrophilic substitution. The 2,4-dicarbonyl motif undergoes cyclocondensation with diamines to form bicyclic heterocycles. Glutarimide derivatives have been shown to inhibit protein phosphatases and cyclin-dependent kinases.

Application

2,4-Piperadinedione is used as an intermediate in the synthesis of glutarimide-based pharmaceuticals including the clinical candidate 6-methylenetricyclene derivatives, thalidomide analogs, and protein phosphatase inhibitors. The piperidine-2,4-dione scaffold is a key structural motif in CDK (cyclin-dependent kinase) inhibitor programs and in the synthesis of angiogenesis inhibitors. It is also used in the preparation of chiral catalysts.
