General Description

Amphetamine sulfate is the sulfate salt of amphetamine, a synthetic central nervous system stimulant belonging to the phenethylamine class. Approved by the FDA in 1976, it is classified as a Schedule II controlled substance with a high potential for abuse, misuse, and addiction requiring careful patient selection and monitoring.

Mechanism of Action

Amphetamine exerts its effects by increasing synaptic concentrations of norepinephrine and dopamine through inhibition of reuptake and promotion of release from presynaptic nerve terminals. As a non-catecholamine sympathomimetic amine, it produces central nervous system stimulation, peripheral elevations of systolic and diastolic blood pressure, and weak bronchodilator and respiratory stimulant actions.

Application

Amphetamine sulfate is FDA-approved for the treatment of narcolepsy and attention deficit disorder with hyperactivity in children as part of a comprehensive treatment program. It is also indicated as a short-term adjunct in exogenous obesity management for patients refractory to alternative weight reduction therapies.

Grochecki P, et al. investigated the long-term cognitive consequences of adolescent exposure to mephedrone (30 mg/kg) or amphetamine (2.5 mg/kg) in rats. When tested in adulthood, both drug-exposed groups showed deficits in spatial memory and reversal learning in the Barnes maze, although the reversal learning impairments may reflect spatial learning deficits rather than specific changes in cognitive flexibility. Mephedrone, but not amphetamine, produced delayed increases in MMP-9 levels in the prefrontal cortex and hippocampus, along with upregulation of GluN2B-containing NMDA receptors. Mephedrone also altered PSD-95 expression in a region-specific manner (increased in prefrontal cortex, decreased in hippocampus).
